Titre: Modification physico-chimique de matrices polymériques par les procèdes de microencapsulation pour la libération contrôlée du piroxicam

Résumé: In the present research, new formulations based on piroxicam(PRX), active ingredient with anti-inflammatory activity and poorly water soluble molecule, have been prepared by the microencapsulation by solvent evaporation process. Microspheres were elaborated using pure PRX and polymeric matrices (EC, EC/HPMC) at different compositions and stirring speeds. In addition, the main objective was to enhance the drug solubility and at once to control the drug release. So, inclusion complexes of PRX-β-cyclodextrin were prepared by dissolution-solvent evaporation and by dispersion and therefore encapsulated in EC and EC/HPMC mixture. The obtained formulations of pure PRX or modified PRX were characterized using different techniques (XRD, FTIR, optical spectroscopy and scanning electron microscopy). Various release profiles were obtained and the release mechanism was identified by modelling using Higuchi and Korsmeyer-Peppas models. The results are remarkably promising and in addition, using the methodology of design of experiments, the effect of some process variables such as the polymer matrix and stirring speed was quantified and evaluated by modelling using the Minitab software 16.
